This study makes two important contributions to the transcription field. First, showing the specificity of RNAPII-CTD kinases CDK9 and CDK12. Second, describing the activation mechanism of CDK12/13 in cells by the PAF1C subunit CDC73. 

A beautiful example of the powerful combination of reconstitution biochemistry, protein folding prediction by AlphaFold and deep sequencing to uncover the mechanism of transcription in humans.
